I did this the last time I got an iphone, it was surprisingly quick. You can do your initial backup at the apple store and icloud will incrementally backup the stuff you add along the way.

They are also the people that made itunes... I needed to wipe my iphone as part of unlocking it, and I did a full wired back / restore. Sort of... it turns out itunes doesn't backup any music you didn't buy through itunes...
