I've left the phone sitting there and iTunes sitting there running for a couple hours and it hasn't done an automatic backup yet, despite showing as connected over WiFi.

So perhaps the words "Automatically Back Up" on that screen in iTunes are a little misleading. Maybe you have to press the "Back Up Now" button. Too bad.
