Will adding an Exchange server to an iPhone that previously didn't have one configured wipe the contacts that currently exist on the phone?

If you choose to sync contacts, yes. If you just need to access email or calendar items, you can do that without wiping the contacts.

Thankfully the backup/restore bit of iTunes/iPhone seems to be reasonably reliable.

If you have MobileMe, the contacts synced via the service won't be wiped.
